There are many approved microsphere-based “depot” products in the marketplace, accounting for billions of dollars of annual sales. Most are based on biodegradable poly(lactide-co-glycolide) (PLGA) microspheres and are administered subcutaneously or intramuscularly with a dosing frequency from once a week to once every several months.
In addition to PLGA, other biodegradable polymers such as polylactide (PLA), polycaprolactone (PCL), polyanhydrides and poly(ortho esters) may also be used as microsphere matrix materials. The release profile of a microsphere product can be controlled by choosing different types of polymers or varying the lactide/glycolide (L/G) ratio in the PLGA polymer.
